Oh yes it does, I just tried it, I used:
C:> SET
(response)
PATH=C:\DOS;C:\DOS\XTREE;C:\WINDOWS

C:> SET PATH=C:\TEMP;%PATH%
C:> SET
(response)
PATH=C:\TEMP;%PATH%
