No. A layer is made of a flat material. It is possible to draw there where should be material and cut outs there material should be removed. Gerber files work in a similar fashion.

Plate thru does however not really follow this model but if there really is a need to draw second material on same layer I guess the most appropriate would be an extra layer at same position/group.
