No, it does not need to be there, and it cannot be used under Windows.

It should not interfere.  EMM386 does have some bugs and limitations (on 
large memory machines for example) which might cause you to not want it
in many situations.

Optional, not required.  You can use raw mode with VDISK if desired.

It depends a lot on the version of Windows you are running.  Hardware
interrupt handling, support for some BIOS and memory functions, long file
names are the biggest differences.
