Make sure that the background colour is #0. This usually involves creating your
own palette. Paint Shop Pro can make a pretty well optimized 256 palette, and
you can edit it in case your transparent colour doesn't come out to what you
wanted it to be. If this happens, modify the colours switching the colours you
need, save the palette only, and cancel your changes to the image. Load it
again, load your modified palette and choose "nearest colour".
