Check your spelling (stdio/studio,djgpp/djpp/digpp).
A small test leading to the error you get:
C:\DC\YR>set DJGPP=e:/mcc/dgpp.env
C:\DC\YR>acd cp
E:\CP>gcc -Wall rot13.c
rot13.c:1: stdio.h: No such file or directory (ENOENT)
E:\CP>set DJGPP=e:/mcc/djgpp.env
E:\CP>gcc -Wall rot13.c
E:\CP>
