No.  Actually, both DOS and Win32 file-related calls go through the
same protected-mode Int 21h dispatch code.  However, it could be that
the Win32 API calls do not use the special bits required for large
file support.  If they don't use those bits, they won't get large
files.

Or, it could be a Microsoft docs inaccuracy (so what else is new?).
