I have ported Web2C-7.3.3 to Cygwin by porting teTeX to Cygwin:
so if you want to use TeX and its friends, under Cygwin, just install teTeX:
do not forget to read the installation doc.
If any trouble, please search in the archive.
To respond to your request:
you have to install a canonical TeXMF structure before to build Web2C.
For more details, download the cygwin source tarball
and read the document relative to Cygwin:
all the stuff I used to built teTeX under Cygwin is there.
